Malarone is an effective anti-malarial tablet. Anti-malarials are necessary when travelling to places abroad where mosquitoes are prominent as the chance of being bitten and contracting malaria is high. These include mainly tropical countries such as parts of Africa, Asia and South America.

What is Malaria and how does Malarone work?

Malaria is caused by numerous types of parasites known as Plasmodium. When bitten by an infected mosquito, the Plasmodium within the mosquito is injected into the bloodstream. Once bitten, the Plasmodium reproduces, resulting in the Malaria.

Proguanil hydrochloride stops the parasite from reproducing in the bloodstream by blocking the enzyme dihydrofolate reductase that is found in the Plasmodium parasite, which is involved in the reproduction of the parasite.

Dihydrofolate reductase converts folic acid into folinic acid in the parasite, which is a necessary step for the parasite to produce new DNA. New DNA is essential for the parasite to reproduce. By blocking this process, proguanil prevents any malarial parasites in the blood from reproducing and increasing in number.

Atovaquone works in a similar way, which is why the use of Proguanil with atovaquone is indicated. Together they are used to prevent malaria caused by a type of Plasmodium called Plasmodium falciparum which produces the most serious form of malaria.

Atovaquone Proguanil may be particularly useful for preventing malaria when travelling to areas where Plasmodium is known to be resistant to other antimalarial drugs, such as chloroquine.

What does each Malarone tablet contain?

The usual tablet dose is 100mg Proguanil/250mg Atovaquone but for children its 25 mg of proguanil hydrochloride and 62.5 mg of atovaquone.

How to take Malarone:

To treat malaria:

  • The usual dose for adults is 4 tablets once a day for 3 days
  • Not for children, talk to your doctor. Not recommended for preventing malaria in children, or in adults who weigh less than 40 kgs

To prevent malaria in adults:

  • Start taking Malarone 1 to 2 days before travelling to an area which has malaria
  • Continue taking it every day during your stay
  • Continue taking it for another 7 days after your return to a malaria-free area

For children the dose depends on their bodyweight:

  • 11-20 kg ? 1 tablet once a day for 3 days
  • 21-30 kg ? 2 tablets once a day for 3 days
  • 31-40 kg ? 3 tablets once a day for 3 days
  • Over 40 kg ? dose as for adults
  • Not recommended for treating malaria in children who weigh less than 11 kgs

For children who weigh less than 11 kgs talk to your doctor. There may be a different type of Malarone tablet available in your country.
