Glucosamine is a natural substance located within joint capsules. In humans, glucosamine is made by chondrocyte cells within joints and used to createglycosaminoglycans and proteoglycans. These by-products of glucosamine merge with collagen to form and maintain healthy cartilage. Glucosamine, glycosaminoglycans and proteoglycans also maintain joint fluid viscosity, which allows for normal lubrication and cushioning, particularly in weight-bearing joints.

Glucosamine & Chondroitin may help prevent knee pain
A large 2006 study conducted by Clegg et al titled 'Glucosamine, Chondroitin Sulphate, and the Two in Combination for Painful Knee Osteoarthritis'and published in the "The New England Journal of Medicine" reported that "Glucosamine and chondroitin sulphate alone or in combination did not reduce pain effectively in the overall group of patients with osteoarthritis of the knee. Exploratory analyses suggest that
the combination of glucosamine and chondroitin sulphate may be effective in the subgroup of patients with moderate-to-severe knee pain".
A 2010 review published in "Rheumatology International" conducted by Lee et al titled 'Effect of glucosamine or chondroitin sulphate on the osteoarthritis progression: a meta-analysis' showed that both glucosamine and chondroitin slowed the progression of osteoarthritis, but only after two to three years of use.
The aim of this study was to assess the structural efficacies of daily glucosamine sulphate and chondroitin sulphate in patients with knee osteoarthritis (OA). The authors surveyed randomized controlled studies that examined the effects of long-term daily glucosamine sulphate and chondroitin sulphate on joint space narrowing (JSN) in knee OA patients.
Six studies involving 1,502 cases were included in this analysis, which consisted of two studies on glucosamine sulphate and four studies on chondroitin sulphate. After 3 years of treatment,
glucosamine sulphate revealed a small to moderate protective effect on minimum joint space narrowing. The same was observed for chondroitin sulphate, which had a small but significant protective effect on minimum joint space narrowing after 2 years.
This meta-analysis of available data shows that
glucosamine and chondroitin sulphate may delay radiological progression of OA of the knee after daily administration for over 2 or 3 years.
